Abstract
The study of DNA structure is necessary for understanding the genetic mechanisms of various cellular processes. Fluorescent dyes (MitoRed, DAPI, Nile Red, SYBR Gold, SYBR Green, and others) are finding increasing application in various fields of cellular and molecular biology. In this study, the effects of different concentrations of fluorescent dyes, specifically 9-nitro-3-(pyrrolidin-1-yl)-7H-benzo[de]anthracen-7-one and 9-nitro-3-(piper-idin-1-yl)-7H-benzo[de]anthracen-7-one, on the UV absorption spectra of double-stranded DNA and oligonucleotides were analyzed to understand the molecular interactions between dsDNS and the novel fluorescent dyes. The results of this study revealed different absorp-tion bands and transitions (bathochromic and hyperchromic shifts) in the spectra of dsDNA and oligonucleotides upon interaction with various concentrations of fluorescent dyes. The obtained data may be crucial for developing novel fluorescent dyes to improve diagnos-tic accuracy based on interactions with dsDNA and oligonucleotides, thereby contributing to a deeper understanding of these processes within biophysics and molecular biology. The obtained data may be of crucial importance for assessing the further application of these dyes for dsDNA labeling and for developing new fluorescent dyes to improve diag-nostic accuracy based on their interactions with dsDNA, thereby contributing to a deeper understanding of these processes within biophysics and molecular biology.
